چکیده

Pain is sensed by the activation of painful nociceptors in periphery or pain mediators, such as bradykinin, serotonin, histamine, and prostaglandin, released from damaged tissue, afferent transmission to medulla spinalis, transmission stages high centers over dorsal horn. Pain, which was seen only a warning sign past, now accepted phenomenon itself that needs be treated search for new, stronger active substances with fewer side effects treatment demand. Hydrogen sulfide (H2S) modulator T-type Ca+2 channels, especially Cav3.2, are known play critical role processing pain. H2S can also show an anti-nociceptive effect opening K+ channels blocking nociceptors. Exciting preclinical data has demonstrated H2S-derived Non-steroidal antiinflammatory drugs (NSAIDs) analgesic agents used treat various types increases resistance gastric mucosa against injury occurred relief accelerates its repair, so it provides advantage derivatized drugs. In addition, donors have been shown induce analgesia through μ-opioid receptors. Based on studies, thought combination opioid receptor agonists may provide additive even synergistic effect. It estimated modification H2S, currently prevent increase effects, promising wise approach.

TRPA1 Has a Key Role in the Somatic Pro-Nociceptive Actions of Hydrogen Sulfide

Hydrogen sulfide (H(2)S), which is produced endogenously from L-cysteine, is an irritant with pro-nociceptive actions. We have used measurements of intracellular calcium concentration, electrophysiology and behavioral measurements to show that the somatic pronociceptive actions of H(2)S require TRPA1. A H(2)S donor, NaHS, activated TRPA1 expressed in CHO cells and stimulated DRG neurons isolate...

Importance of anti- and pro-nociceptive mechanisms in human disease.

T he burden of chronic pain to society is enormous. This is both in terms of physical and emotional impact to individuals and carers, in addition to the large financial burden. Current estimates suggest that 11.5–55.2% of individuals worldwide are defined as suffering from chronic widespread pain. A major characteristic of functional disorders such as irritable bowel syndrome (IBS) and inflamma...

The neurobiology of nociceptive and anti-nociceptive systems.

Pain is both a sensory event of the peripheral and central nervous systems and an experience that arises from, and reciprocally can affect processes of higher consciousness. Alterations in peripheral and central neuraxes can lead to sensitization, structural modification and long-term potentiation that can overcome modulatory influence(s) and change the non-linear adaptability of these systems ...

Anti-Nociceptive and Anti-Inflammatory Activity of Araucaria bidwillii Hook

The effect of alcoholic extracts of leaf from Araucaria bidwillii Hook. [ABH] (Family: Araucariaceae) was evaluated in experimental models of pain and inflammation. Oral administration of 100, 200 and 300 mg/kg of leaf extracts of ABH were used for the above study. The leaf extract at 300 and 200 mg/kg showed significant reduction in acetic acid induced writhings in mice with a maximum effect o...

Receptor Mechanisms Mediating the Pro-Nociceptive Action of Hydrogen Sulfide in Rat Trigeminal Neurons and Meningeal Afferents

Hydrogen sulfide (H2S), a well-established member of the gasotransmitter family, is involved in a variety of physiological functions, including pro-nociceptive action in the sensory system. Although several reports have shown that H2S activates sensory neurons, the molecular targets of H2S action in trigeminal (TG) nociception, implicated in migraine, remains controversial. In this study, using...
